Obiettivi formativi

<br />Fornire agli studenti il concetto che "L’INGEGNERIA GENETICA E’ SOLAMENTE UNA CONSEGUENZA DELLA RICERCA DI BASE. MOLTE DELLE PROCEDURE SONO NATE COME CONSEGUENZE IMPREVISTE DI ESPERIMENTI ESEGUITI PER SODDISFARE LE CURIOSITA’ DI QUALCUNO SULLANATURA."(A.Kornberg)<br /><br /><br /><br /><br /> To provide the students with the concepts that bacterial genetics has probably had more profound impact on the development of modern molecular biology : Recombinant DNA techniques arose directly out of research in bacterial and bacteriophage genetics. Moreover , it is interesting to known how phenomena were discovered, how the problems were born, attacked and solved, and how and why our ideas have evolved.

Contenuti dell'insegnamento

<br />In questo corso si metteranno a fuoco le relazioni tra il campo della genetica e quello della batteriologia , che convergendo tra loro hanno influenzato lo sviluppo della biologia moderna.<br />Il corso sara’ presentato secondo la sequenza temporale con la quale la materia e’ stata effettivamente scoperta, e presentera’ i fondamenti della genetica molecolare, cosi’ come essi sono stati via via scoperti descrivendo almeno una parte del materiale sperimentale.<br />Saranno cosi’ descritti gli esperimenti cruciali, compiuti negli anni quaranta e cinquanta,che costituiscono il punto di riferimento per lo sviluppo della genetica batterica e che hanno permesso lo sviluppo delle tecnologie del DNA ricombinante:<br />1. L’isolamento da parte di George Beadle and Edward l. Tatum (1945)dei mutanti nutrizionali nel fungo Neurospora crassa. Gli esperimenti di Beadle/Tatum portarono al concetto di "un gene/un enzima" : la prima reale idea riguardo alla connessione tra genotipo e fenotipo.<br />2. L’esperimento di Luria/Delbruck(1943),successivamente conosciuto come "Test di fluttuazione", che ha messo le basi per gli studi quantitativi sulla mutazione batterica.<br />3. La dimostrazione da parte di Avery,MacLeod and McCarty(1944) che il principio trasformante dello pneumococco e’ il DNA e l’esperimento di Hershey and Chase(1952) che ha dimostrato che e’ il DNA del batteriofago che entra nella cellula durante l’infezione.<br />4.La scoperta del sesso in Escherichia coli K-12 da parte di Lederberg e Tatum(1946).<br />5.La dimostrazione che il batteriofago e’ capace di subire la ricombinazione genetica (Delbruck and Bailey,1947; Hershey,1946).<br />6.La dimostrazione da parte di Lwoff and Gutmann (1950) che la produzione di fago da batteri lisogeni e’ un evento cellulare piu’ che un evento di popolazione.<br /><br /><br /><br /><br /> In the present course , the key research findings of bacterial genetics that led to the present era of molecular genetics have been brought together. I am interested here primarily in the experimental work that drove bacterial genetics forward. Althoug scientific wisdom comes from the empiricism of experimental research, raw data are of little interest for their own sake. It is only when these data are interpreted in the light of an intelligent model that the advancement of scientifc knowledge and understandind occurs.<br />This course is a history of science rather than of scientists and is concerned primarily with 20th century science. The key experiments (listed below) that can be said to constitute a turning point in the developmment of bacterial genetics all came within a few years of each other in the 1940s.<br />1)     The isolation by George Beadle and Edward L. Tatum of nutritional (biochemical) mutants in the fungus Neurospora crassa , making possible the study of rare genetic events because of the power of nutritional selection. The Beadle /Tatum experiments also led to the one-gene/one-enzyme concept, the first real insight into the connection between genotype and phenotype( Beadle, 1945).<br />2)     The Luria/Delbruck experiment, subsequently known as the fluctuation test, which became the basis for quantitative studies on bacterial mutation (luria and Delbruck, 1943).<br />3)     the demonstration by Avery, MacLeod, and McCarty (1944) that the transforming principle of pneumococcus was DNA, and the experiment of Hershey and Chase(1952) showing that the DNA of the bacteriophage that entered the cell during infection.<br />4)     The discovery of mating (sex) in Escherichia coli K-12 by Lederberg and Tatum (1946).<br />5)     The demonstration that bacteriophage is capable of undergoing genetic recombination (Delbruck ,1946; Hershey,1946).<br />6)     The demonstration by Lwoff and Gutmann (1950) that phage production by lysogenic bacteria is a cellular event rather than a population event.<br /> <br />
